The HIV Proviral DNA test is 99.9% accurate from 10 days post exposure
Proviral DNA is created when the HIV virus enters a cell and uses the reverse transcriptase to make DNA copies of the RNA genome. These copies, called proviral DNA, are either found in the cytoplasm of the cell or become part of the host cell.
A very simplified explanation of how the test is preformed is peripheral mononuclear cells (PBMC mainly lymphocytes) are lysed and the genetic material is than amplified through a replication process which is repeated 20 to 30 times. Through the amplification, the selected region may be amplified more than a million times. Proviral DNA may be detected in both cells that are actively replicating virus as well as those cells that are only harboring the proviral DNA.
